I'm planning on hitting NYC in October, will be staying Downtown (Wall Street area) looking for tips on good spots in the City to check out.
I am very familiar with NYC and would not be interested in touristy spots, just the nightlife related establishments from restaurants, lounges to bars/clubs.
Any good places to find some dimes?

Meatpacking district is where all the new hot clubs/lounges are. Gans a Voort and Gaslight are typically good. M2 if you're a masochist.
East Village if you want some good dives and fun watering holes. I like Solas and Porch.

Ulysses/Stone Street on Thursday night, go early, the scene peaks at 10 or 11. This is in the Wall Street area so you can walk there.
The Standard Biergarten, Brass Monkey, Gaslamp, and Bar 675 are the no nonsense/no door-bs places in the Meatpacking district, where the hottest girls will be. I prefer them in the order listed. Son Cubano will require some door finesse but it has a lot of latino girls. Gansevoort is beyond cheesy now. Tenjune is still hot (difficult door), as is One. The Standard's Boom Boom room is the toughest door in NYC right now but the talent is off the charts.
M2 in chelsea is now closed. Rightly. West Chelsea is where you will find the megaclubs but I haven't done that in years.
Agree with Entropy on Solas and Porch -- I've had one night stands at both recently, the girls are open. Additional decent East Village spots: Niagara, Horseshoe Bar, Sunburnt Cow, International Bar (very divey), B-Bar, Double Diamond (the back room), Von's, D.B.A., No Malice Palace.
As for the lower east side, Ludlow and Essex streets between Houston and Rivington becomes a massive carnival of people and women on weekend nights, there are about 20 bars there, choose one (Pianos is the classic (bar downstairs, dancing upstairs)). This area will be happening on Thursday night as well.
Lily's off of Union Square is also good and one of my current low key favorites. The Globe on 23rd draws a goodlooking crowd of girls but usually young and cliquish (there is dancing there, too).
The club downstairs from the Ace Hotel is a current hotspot, I went once and it was very good. Enter through the Breslin restaurant, a good bar scene in its own right, wait on the list to get into the library -- another good bar scene, then go downstairs to the "theater." A new Gansevoort hotel has just opened up near it, have not been there, sure it is sceney.
If you find yourself in Brooklyn, Brooklyn bowl is awesome, a bowling alley slash concert venue.

I just had a conversation about this with one of my friends from NYC.
If I recall, he mentioned:
Lavo NY
The Darby
Don Hill's (I think this is a live music joint, so it might be crap)
The Standard's Boom Boom room
I bet they are all rough doors.
I think he also mentioned a new W Hotel opened (I think it is downtown actually), which I am sure will have a scene for a bit, but don't ask me, I don't do W Hotels. But this could be your move.
Flashman, good data sheet.
Update: I think Rose Bar in Gramercy Park Hotel is still ill.
I haven't been to NYC in a while.

I'd like to recommend The Pig and Whistle if it's still open.
I think it was on Lexington a few blocks over from the Four Seasons Hotel, which also had a great bar in it. The Pig and Whistle always had great bands and a real good selection of girls. There was an old lady bartender that was a sweetheart. She'd break her ass working on the really busy nights and was cool to watch work.
Aloha!

Gracias for all the responses. I'll report back on the trip.
On a related note, I'll be going suited down. Not concerned about the meatpacking district but LES is a question mark with the hipsters. I don't want to be mistaken for a wall street dbag (nothing against the street, just hate the guido turned into stockbroker with sharkskin suits stereortype).
Fortunately I know my way around the city and would have no problem posting up with some street game as well.

CAPITALE.
During a city trip, we attended a party there by chance. Didn't know what to expect, and I was absolutely blown away.

If you have an iPhone I'd suggest you get this dope app called NightBox that really gives you the 411 for the nicest and trendiest spots in the city. The NY nightlife market is brutal and many places that were hot 12-16 months ago suck now. Another thing you have to worry about is actually getting inside. At the best places unless you know an owner, promoter or the door person you probably don't have a chance of getting in. There are many venues that won't even let in groups of girls that don't have the right connection.
With that said though the nightlife in NY is almost unrivaled in the world. You can probably count on one hand the number of cities in the world that have the quantity and quality of lounges/clubs that NY has.
Oh, and if there is anyone out there who has connections to get inside of The Top of the Standard (aka Boom Boom Room) PLEASE hit me up with a message.

Highly recommend the Soho Grand. Bonus is active nightlife and hotties in the bar downstairs so you may not have to venture far to find fly girls.
Not quite in Soho, but the Bowery Hotel is around the corner in the East Village. Location is great, right at the nexus of the EV and the LES. Dim lighting, possibly quieter than the Grand but in a more bustling area. Nice and discreet service. Big fireplace in winter. Less of a party place than the Grand.
Those two rule the roost for luxury hotels in that part of the city as far as I'm concerned.
I've heard good things about the Thompson too but have no first-hand experience of it. Seems to have less character than the above.

I would definitely spend some time in the west village around Washington Square Park and NYU.
Don Hill's which G manifesto mentioned is dead now. It used to be good when my cuz was the DJ for the weekly Thursday party "frankie jackson's soul kitchen".

I'm not a New Yorker and not too familiar with the city, but I've met hot girls at Juliett, Green House and the Box. I don't know how those places compare to other clubs in NYC but I did get lucky at Juliett (not SNL). My local friend took me to those places and I'm not sure if it's easy to get in without a hookup.
