I just returned from Santo Domingo where I spent a few days with my crew. It was awesome. There's already many good data sheets on the place (Elabayarde, Lumiere, etc) so I won't repeat too much of what has already been said.
Lodging
Definitely stay in a hotel or an apartment, but make sure you know up front what the deal is with the visitors policy. We rsvp'd at the Renaissance which allows guests, but when we showed up, they said that you can only have two visitors per guest and their names need to be provided upon check-in only, not later. This may work if you're going down to see girl you already have lined up but not if you're going to hunt. We immediately cancelled the reservation and switched to a Marriott on Maximo Gomez which was even better actually and they didn't care if you brought in 20 girls. If you're going to do an apartment try to get one in the Zona Colonial area. There's a few good bars and restaurants there and it's generally safe.
Transportation
There's public transport in SD (metro trains, buses), but the best way to get around is by taxi. You can also rent a car but I don't recommend it as the way people drive in SD is batshit crazy and chaotic. It's actually better to just hire a driver for the day which can be even cheaper than renting a car. Gas in the DR is like $5/gallon.
Girls
There are tons of girls in Santo Domingo. Like everywhere. This is one thing that surprised me greatly. Between Brazil and Colombia, I thought I had Latin American women all figured out as far as looks were concerned. Boy, was I wrong. DR now belongs on my rotation with BR and CO. I actually plan on returning to the country before the end of the year to visit the north coast and Cibao Valley (home to the best looking girls in the country). You are truly losing if you neglect to visit this place. Every single night we went out we met up with new girls. They are very easy to hook and are either pay for play or just sluts who enjoy fucking. Otherwise, the high end and better educated girls are more long-term projects (relatively speaking) so that's good if you have some time to see them again. Like Lumiere said in his data sheet, verify up front whether the girl you have is p4p or not. It's not easy to tell all the time because they mingle and dance in clubs like normal girls.
Also, the thing to keep in mind is that girls in DR, p4p or not, want something out of you if you're a gringo. They want you to buy drinks or something. The culture of DR is very much like that. Everyone is looking out for the advantage.
Speaking Spanish will still help you greatly. Dominican spanish is a much different dialect than other kinds. It's spoken faster, enunciated differently, and has a lot of slang.
As for spots to check out, there are several options. Lets start with the Zona Colonial area where most of the action is centered around Parque Colon. I like Onno where they play hip-hop and American/European music (my favorite venue personally). There's also La Cantina de El Agave for some Dominican music (Merengue/Bachata) and reggaeton. This is where you come to dance which is very important in DR culture. There's also some bars like Segafredo and the Hard Rock cafe. When you walk farther away from the park you will find isolated bars and clubs scattered throughout the Zona Colonial which could be hit or miss.
Zona Colonial can not be neglected for day game. I would say do that at least as much if not more than night game. This area has many malls and shopping on pedestrian only streets as well as sidewalk cafes. I often used the "excuse me, do you speak English?" approach which immediately displayed my foreigner status which is what you want to go for in DR. I then switched it up to Spanish. I never got outright rejected going this route. When girls spoke English, they said they did and would be receptive to you. If they didn't speak English they would usually say sorry and keep it moving.
Another area for nightlife is Avenida Venezuela which is on the other side of town (across the bridge) in an area called Santo Domingo Este (East Santo Domingo). The girls here are the best I've ever seen in the city. Ridiculous and relentless talent. This is a local scene however and every club and bar plays strictly bachata and merengue music. The venues are also very loud so it makes for approaching very tedious. These are the kind of places where dance game would be your best bet, but you have to know bachata and merengue somewhat. Another option is to cold approach on the street, but these girls usually only speak Spanish.
There's some other spots on this side of town. I would defer to RVF member Elabayarde for this intel.
Back on the west side, in addition to Zona Colonial, you also have clubs/casinos that are part of hotels along the Malecon or George Washington Avenue. Renaissance, Napolitano, Intercontinental, etc. The clubs Mint and Pure are worth pointing out in this area. The music is local Dominican bachata and merengue stuff.
All in all, if you go out and approach, you will either get laid or come very close to getting laid every night in Santo Domingo. This is a very fun city to go with your boys. A true "mancation" destination. It seems like the whole city is setup for a man to enjoy. Anything you want, they got it.
I also recommend a day trip or overnight stay at one of the beaches in the area, either Juan Dolio or Boca Chica. They are within an hour from Santo Domingo and you can have a good time, whether you bring a chick with you or are looking for one. They've also got beach service down to a science. Anything you want just right on the beach.

Picked me up with his driver and I introduced him to some girls in my crew at the club I was at. Everyone else, DR is probably one of the easiest places to get laid in Latin America. The island has a very sensual vibe and the chicks are sexy as hell. They are also cool and easygoing to talk to. For Americans, this is also a relatively easy part of the world to get to. It's a trip that can be done multiple times a year which is what I plan to do now for DR. I think it offers the most bang for the buck in Latin America. Unlike Brazil, DR is relatively cheap and it's easier to get to (only a 3-4 hour direct flight from NYC or DC, and 1 1/2 from Miami) which is at least half the amount of time it takes to get to Brazil. Also, the cultural barrier isn't nearly as great. American style capitalism has infiltrated DR pretty hard. You'll find TGI Fridays, all of the American fast food places, etc. Even if you don't speak Spanish, getting along in DR won't be a problem. My boys who didn't speak Spanish enjoyed the trip greatly. Colombia is a great country but the girls aren't as easy and Spanish is almost necessary to really enjoy it.
