I have visited Brno for less than a week for a conference.
The expectations were high as this is 2nd tier city in EE with large university population - 90,000 students for a city of ~400,000 people.
The city itself is pleasant with beautiful old center, good food and great beers by local breweries.
Girls:
Coming from Poland, Czech girls were disappointing, average talent was less and quite a few were not taking care of themselves, even saw some girls with cute faces and good bodies which has things like yellow teeth and dirt under nails...
But still the average is much better than most western countries and you can spot a lot of 7s all over.
Exotic factor is not high due to that among the 90,000 students many are foreigners and they go out a lot, add to that low cost flight so in the bars and clubs all the time many foreigners from many countries.
You will have better exotic factor with girls which are none-students or coming from more rural area, English would be less good in this case.
On the advantage I found Czech girls more liberal, had 1 SNL and some heavy sex dances and saw around people hookup.
I met quite a few Slovak girls there, in general they were more educated and more warm than Czech but this could be due to that they are traveling girls, they did improve my opinion on Slovak as in the past I was in nearby Bratislava and found did quite westernized.
Where to stay:
In the old center, no doubt about it, everything is happening there, nightlife, day game, universities are nearby. All of it is walking distance, in this term this city has good logistics.

If I had now the choice, for Czech I would go to Ostrava/ Olumuch as those cities are less hit by foreigners. That said, Brno is by no means Prague and one can pull there with good game and if staying for a long term via social circle.
